Retinal Ganglion Cell Type, Size, and Spacing Can Be Specified Independent of Homotypic Dendritic Contacts

چکیده
In Brn3b(-/-) mice, where 80% of retinal ganglion cells degenerate early in development, the remaining 20% include most or all ganglion cell types. Cells of the same type cover the retinal surface evenly but tile it incompletely, indicating that a regular mosaic and normal dendritic field size can be maintained in the absence of contact among homotypic cells. In Math5(-/-) mice, where only approximately 5% of ganglion cells are formed, the dendritic arbors of at least two types among the residual ganglion cells are indistinguishable from normal in shape and size, even though throughout development they are separated by millimeters from the nearest neighboring ganglion cell of the same type. It appears that the primary phenotype of retinal ganglion cells can develop without homotypic contact; dendritic repulsion may be an end-stage mechanism that fine-tunes the dendritic arbors for more efficient coverage of the retinal surface.

Do-It-Yourself Tiling Dendritic Growth in the Absence of Homotypic Contacts
Many subtypes of neurons are precisely arranged with little overlap of their dendritic arbors, which has been suggested to arise due to homotypic dendritic repulsion. In this issue of Neuron, Lin et al. demonstrate that subtypes of retinal ganglion cells establish normal dendritic field sizes and morphologies in the absence of contact with cells of the same class.
متن کاملRetinal Mosaics: Pattern Formation Driven by Local Interactions between Homotypic Neighbors
Individual types of retinal neurons are distributed across the retina as regular arrays of cells, commonly referred to as “retinal mosaics” (Wässle and Riemann, 1978). This regularity in their patterning is generally assumed to ensure that all locations across the retina are sub-served by every type of retinal neuron, each participating in their own unique way to the local computations carried ...
متن کاملAfferents and homotypic neighbors regulate horizontal cell morphology, connectivity, and retinal coverage.
Horizontal cells are inhibitory interneurons with laterally oriented dendrites that overlap one another, contacting the pedicles of cone photoreceptors. Because of their regular spacing, the network of horizontal cells provides a uniform coverage of the retinal surface. The developmental processes establishing these network properties are undefined, but cell-intrinsic instructions and interacti...
متن کاملA Mosaic of Synaptic Contacts among Three Retinal Neurons
Retinal bipolar, amacrine and ganglion cells contact each other within precisely defined synaptic laminae, but the spatial distribution of contacts between the cells is generally treated as random. Here, we show that not to be the case. Excitatory inputs to inner retinal neurons were visualized by introduction of a plasmid coding for the postsynaptic protein PSD95-GFP. Our initial finding was t...
متن کاملRetinal Ganglion Cell Complex in Alzheimer Disease: Comparing Ganglion Cell Complex and Central Macular Thickness in Alzheimer Disease and Healthy Subjects Using Spectral Domain-Optical Coherence Tomography
Introduction: Alzheimer disease (AD) is the most common form of dementia worldwide. The modalities to diagnose AD are generally expensive and limited. Both the central nervous system (CNS) and the retina are derived from the cranial neural crest; therefore, changes in retinal layers may reflect changes in the CNS tissue. Optical coherence tomography (OCT) machine can show delicate retinal layer...
